This is a surface display image of BCL-2, bound with venetoclax. The style chosen for this image highlights the binding pockets, making them easier to visualize. However, it also obscures some of the alpha helical secondary structure. I plan to modify individual key residues that have been shown to produce resistance mutations. I aim to display these modifications using nanome, which should be possible. Unfortunately, pymol does not offer a modify option, and its surface shape displays make it difficult to discern individual residues. My goal is to alter the shapes of the binding pockets with simple one-residue mutations. The function of the linear ligand on the back side, labeled or nonaethylene glycol, remains undetermined at this point. For now, I will treat it as an artefact until its function can be conclusively established. The research reference for this study is DOI: 10.1038/s41467-019-10363-1.
