mTORC1 Signalling Plays a Vital Role in Cell Proliferation and is Essential for Cellular Processes. Defects in this System can Lead to Aging, Chronic Diseases, Cancers, and Other Health Issues. The G-Protein Rheb, a Master Regulator Bound with GTP, Activates mTORC1. Through Mutation Studies, it was Discovered that Rheb Binds with mTORC1 Through the Switch II Region. The Molecule NR1, in Yellow, Inhibits mTORC1 Signalling by Blocking and Decreasing the Affinity of mTORC1 Binding to Rheb, Interfering with its Signalling Process. With an IC50 of 2.1 μM, NR1 Exhibits High Potency. Unlike Other Proposed Drugs such as Rapamycin, which Inhibits mTORC2 Signalling and Causes Adverse Effects, NR1 is a Unique and Selective Inhibitor of mTORC1 Activation, Irrespective of Treatment Duration.
