Replication- Gene Synthesizer by Kong, Jacobson

A four-chambered microfluidic chip is fabricated using PDMS and glass for DNA synthesis purposes. The device features water-cooling jackets in each chamber, along with a control layer to maintain optimal reaction temperatures. Stereolithography etched master molds are employed to create the flow and control layers via PDMS casting. With dimensions of 40 x 25 x 2 mm, it includes 100 µm wide input channels for flow and 300 µm wide control channels. This design is inspired by Kong, Jacobson et al.'s research on parallel gene synthesis in a microfluidic device (https://www.ncbi.nlm.nih.gov/pmc/articles/PMC1885655/pdf/gkm121.pdf) and MetaFluidics' Microfluidic Gene Synthesizer (https://metafluidics.org/devices/microfluidic-gene-synthesizer/).
