In this structure, IRS2 binds to the kinase active site with Tyr628 poised for phosphorylation. Although Tyr628 was indeed phosphorylated by the insulin receptor, its turnover rate was remarkably poor, leading to significant kinase inhibition. Our research clearly shows that the KRLB region functions to restrict tyrosine phosphorylation of IRS2.
