PDB: https://www.rcsb.org/structure/3bu3 IUPHAR: http://www.guidetopharmacology.org/GRAC/FamilyDisplayForward?familyId=321\nInsulin receptor substrates 1 and 2, crucial adaptor proteins, mediate insulin's metabolic and mitogenic effects as well as those of insulin-like growth factor 1. Bound in the kinase active site within this structure is a segment of IRS2, where Tyr628 waits for phosphorylation. Despite being phosphorylated by the insulin receptor, Tyr628's catalytic turnover was slow, leading to kinase inhibition. Our research shows that the KRLB region limits tyrosine phosphorylation of IRS2.(PubMed abstract)
