Argonautes (Ago) are a family of proteins that play a crucial role in the small RNA silencing pathway as "slicers." These proteins cut target RNA strands using an siRNA as a guide, forming the main component of the RNA-induced silencing complex. Among humans, four different types of Ago proteins have been identified, but only one, hAgo2, shows slicing activity; the other Ago proteins, despite having very similar structural conformations to hAgo2, do not function as slicers in their natural form. In 2013, a study aimed to clarify the molecular structure of inactive hAgo1 and to identify peptide domains and amino acid residues involved in hAgo2's catalytic activity that have been altered in hAgo1 and other hAgo proteins.
