Lesson Plans and lab work is now available on my teacher pay teacher site, https://www.teacherspayteachers.com/Product/3D-Printable-DNA-RNA-Manipulative-Lab-1-3452084. This is a set of restriction enzymes to expand upon my DNA/RNA Manipulative set. http://www.thingiverse.com/thing:1259352. These 4 restriction enzyme blocks can be used to model how they work, and how gene splicing/recumbent DNA works. The blocks also show how many restriction enzymes are palindromes with sticky ends. Your students will enjoy simulating genetic engineering with this expansion set. This features EcoRI, BamHI, HindIII, and KpnI Restriction Enzymes. There are over 600 of these restriction enzymes used in gene splicing. If you want to make your own restriction enzymes for gene editing, I offer an expansion set here http://www.thingiverse.com/thing:1321230. A partial list of restriction enzymes can be found here, https://en.wikipedia.org/wiki/List_of_restriction_enzyme_cutting_sites. If you want other restriction enzymes made, I can make them on commission. PM me or contact me via stan@3dpt.club for details. Print Settings Printer Brand: LulzBot Printer: TAZ 5 Rafts: Doesn't Matter Supports: Doesn't Matter Resolution: .3-.4 mm Infill: 5% Notes: I printed them in clear filament so they can lay over a DNA sequence and you can find where they match. Break all sharp edges. If you can't see the lettering, use a permanent marker to highlight the edge of the letters. Print 2 of each enzyme so you can model the cutting pattern and cleaved fragments. Post-Printing Clean up After printing break any sharp edges. I found an emery board works great for rough edges. If you print clear, use a permanent marker to highlight the edge of the lettering. How I Designed This Design and printing These were designed in TinkerCAD based on the previous DNA/RNA file. They were designed to fit as one piece so you can see how they work, and save time building them with the other set. If you want to expand and make your own, DL my expansion set here http://www.thingiverse.com/thing:1321230. Custom Section Project: Gene Splicing and Restriction Enzymes This is to expand upon the DNA/RNA manipulative set. Your students will see how gene splicing, and how restriction enzymes are used to splice new genes into an organism. You can visit https://en.wikipedia.org/wiki/List_of_restriction_enzyme_cutting_sites for a list of restriction enzymes and their cutting sites. You can make your own with this expansion set http://www.thingiverse.com/thing:1321230. Objectives Students will understand what genetic modification is. Students will model how restriction enzymes work. Students will model gene splicing. Students will demonstrate how to alter DNA. Students will discuss the ethics and implications of genetic modification. Students will discuss genetically modified organisms (GMO's) and agriculture. Students will show their knowledge of DNA and RNA. Students will model how mecumbant DNA works. Students will show where the sticky ends are. Steps! a. Build a model of DNA matching the restriction enzyme. b. Model how the restriction enzyme cuts the DNA along specific base pairs. c. Show where the sticky ends are. d. Use the enzymes to insert a new DNA sequence. e. Discuss gene splicing. Show organisms that have undergone this procedure. f. Discuss the ethics on gene splicing. Results To demonstrate knowledge of this, have the students work in small groups to demonstrate the process or teach it to others. You can have the students make a stop-motion video of the process of gene splicing. You can also have them show what they have learned by modeling DNA replication, RNA transcription, protein synthesis, and gene splicing to produce new proteins. You can tie it to pop science by having them watch sections of the film Jurassic Park/Jurassic World that discuss what they did to create the dinosaurs. With that have the students model what the scientist in the film did.
