Researchers are zeroing in on a prime target for creating an anti-viral treatment for COVID-19: the main protease of SARS-CoV-2 (Mpro), which contains the RNA genome. The Mpro is essential for transcription and viral replication, as it processes and matures replicase polyproteins through complex biochemical pathways. By blocking the effects of this protease, scientists can prevent the disease from spreading rapidly. A detailed analysis of the crystal structure of COVID-19 virus Mpro in combination with the mechanism-based inhibitor N3 reveals key targets for the inhibitor to bind tightly to the enzyme. The N3 covalent bond with the enzyme shows tremendous potential for deactivating the protease, a critical step in developing anti-viral medications and containing an epidemic's devastating impact.
