GDP is tightly bound to a specific subunit of the G-protein complex. This interaction stabilizes the G-protein in its inactive state and prevents it from interacting with downstream effectors. When an agonist binds to the G-protein coupled receptor, it induces a conformational change that releases the GDP from the G-protein, allowing the GTP to bind instead.
